    Synthetic biology tracker: milestones and policy

    This tracker follows verifiable milestones in synthetic biology and biomanufacturing: genome writing, circuit design methods, DNA synthesis capacity, fermentation plants and screening policy.[1][2][3] As of October 2026 the biggest open items are the pending replacement of the 2024 US nucleic acid synthesis screening framework and whether new pilot plants fill.[4][5]

    Where the field stands

    Three threads move at different speeds. Genome writing has delivered finished artefacts: a completed synthetic yeast chromosome set and a recoded four-megabase bacterial genome.[6][1] Design methodology changed in early 2026, when very large measured libraries plus machine learning replaced hand-tuning for genetic circuits.[2][7] Industrial capacity is the slowest thread, and the one now absorbing public money.[3][8]

    The commercial split is stark. Input suppliers grew: Twist Bioscience reported record revenue of $118.4 million in the quarter to June 2026.[9] Organism-engineering services shrank: Ginkgo Bioworks reported $20.2 million with government and defence customers supplying 45% of revenue.[10][11]

    What to watch next

    The clearest near-term signal is governance: the replacement for the 2024 US synthesis screening framework ordered in May 2025 has not appeared, and its scope will determine whether screening remains a funding condition or becomes something broader.[4][12]

    On capacity, watch opening dates and utilisation at BioMADE‘s sites: BioMADE aimed to open the Hayward pilot plant to customers in early 2026, and its published Iowa date has moved between 2027 and early 2028.[5][13] On products, further GRAS outcomes after the September 2025 fermented egg protein letter will show whether the regulatory path is repeatable.[14]

    On technology, two things would count as real progress: construct lengths beyond today’s 50-kilobase commercial ceiling, and evidence that AI-assisted circuit design transfers from human cell lines into production hosts.[15][16][17] Cell-free routes are worth watching through public funding, including the August 2026 ARPA-H award for cell-free DNA bioproduction.[18]

    Design and measurement

    Two 2026 results changed what counts as a design method. In January, high-throughput mapping measured hundreds of thousands to millions of genetic circuit designs in one experiment, and models trained on those measurements predicted untested variants; all 40 designs checked by hand matched.[2][7] In October, Nature Biotechnology published Shotgun Genetic Engineering, which delivers many barcoded small constructs into mammalian cells instead of one large construct, so each cell tests a different pathway and working combinations are found by sequencing barcodes.[19][20] Its authors screened millions of combinations in CHO and Jurkat cells, reached near-wild-type growth in valine-free medium and, they say for the first time, isoleucine prototrophy in CHO cells, with working solutions needing 23 to 52 kilobases of synthetic DNA.[21] That length requirement ties design throughput back to synthesis capacity.[15]

    Protein design is being pulled into the same loop. On 18 August 2026 Twist Bioscience disclosed in an SEC filing that anthropic had chosen it as one of the independent evaluators to make and test proteins its models had designed against 15 targets.[22] Anthropic reported binders against 14 of the 15 targets, with hit rates of 22.6 to 35.1 percent against the 10 to 15 percent it says is typical, using up to 12,500 NVIDIA H100 hours per run.[23][24] These are the model developer’s own figures, published alongside the filing rather than in a peer-reviewed paper, so they are recorded here as reported.     Why these entries

    The timeline mixes three kinds of event on purpose, because the field advances unevenly. Published results date the science: a completed synthetic chromosome set, a recoded bacterial genome, a new way to measure circuit designs.[6][1][2] Filings and company announcements date the industry, including quarterly DNA volumes and the restructuring of the best-known cell engineering company.[25][26] Government actions date the rules and the money, from the screening framework to pilot-plant investment.[27][3]

    How this tracker is maintained

    Entries are added only with a dated source and are tagged confirmed, reported, disputed or unknown. The modern era here is dated from around 2010, when US screening guidance for providers and users of synthetic nucleic acids was first issued and synthetic genomes began to be installed in cells.[28] Official feeds for this field are listed in the pipeline configuration so new milestones surface automatically.[29]
