LOY-002 (Loyal's senior dog lifespan drug)

LOY-002 is an experimental daily pill from the company Loyal, intended to extend the lifespan of dogs aged 10 and older by improving insulin sensitivity.[1] As of October 2026 the FDA has accepted its effectiveness and safety sections, but manufacturing review remains before conditional approval can be considered, and approval is not guaranteed.[2][3][4]

LOY-002 is a once-daily pill developed by the company Loyal for senior dogs, aged 10 or older and weighing at least 14 pounds. Loyal says it is intended to preserve quality of life and extend lifespan by improving insulin sensitivity and lowering insulin levels.[1] It is a veterinary product, regulated by the FDA’s Center for Veterinary Medicine.[3]

Regulatory path
LOY-002 is following a conditional approval route with three major requirements.[4]
- Effectiveness (RXE). In February 2025 the FDA accepted the reasonable expectation of effectiveness section. A Loyal executive described this as meaning the drug has a reasonable expectation of extending a dog’s life.[2]
- Safety (TAS). In January 2026 the FDA accepted the target animal safety section.[3] Loyal says this was part of an Expanded Conditional Approval (XCA) application, and frames the drug as addressing the metabolic dysfunction that all dogs develop with age.[6]
- Manufacturing. This section must still be accepted before conditional approval can be considered. As of October 2026, Loyal’s website says two of three major requirements are complete and that FDA approval is not guaranteed.[4]
Acceptance of the effectiveness section is based on preliminary data. It is not proof that the drug extends life.[2]
The STAY study
Loyal’s STAY study completed enrolment in July 2025 with about 1,300 dogs at 70 US veterinary clinics.[7] As of October 2026 Loyal’s website gives the exact figure as 1,317 dogs.[8] It is a separate effort from TRIAD, the Dog Aging Project’s randomized trial of rapamycin in pet dogs, which plans to enrol 580 dogs and uses survival time as its primary outcome.[9][10] STAY is roughly twice that size.[8][10]
Loyal’s other programs
LOY-002 is one of three drug programs Loyal lists as in development.[8] The other two target large and giant-breed dogs:
- LOY-001, an injectable drug, received RXE acceptance in November 2023. Loyal describes it as the first drug to achieve an RXE acceptance for lifespan extension in any species.[11]
- LOY-003, a once-daily tablet, received RXE acceptance on 16 September 2026, according to Loyal. Like LOY-001, it is designed to lower growth hormone and IGF-1, hormones that drive growth and are linked to shorter lifespans in big dogs.[12]
The growth and insulin signals these drugs act on are part of nutrient sensing, and deregulated nutrient sensing is one of the twelve hallmarks of aging.[13] All three RXE acceptances rest on preliminary evidence, not completed lifespan trials.[2]
Why it matters
